Note: The heater/shaker is turned on and off using this switch. Note: The front cover is a hinged, transparent cover that provides easy access to the deck. The cover is closed and locked during a run. Note: The auto-load tray extends from the front of the deck with track markings to assist the carrier placement. The tray is divided into 30 tracks along with etched deck symbols. Samples, reagents and consumables are loaded onto the auto-load tray in the carrier at specific track positions. Note: The deck is an enclosed work surface that is divided into 30 tracks. Samples, reagents and consumables are loaded onto the deck in carriers at specific track positions by the auto-loader. Note: The barcode reader automatically scans the carriers, reagents and consumable barcodes during a run for positive identification. The reader rotates to scan both horizontal and vertical barcodes. Barcode Reader The barcode reader automatically scans the carriers, reagents and consumable barcodes during a run for positive identification. The reader rotates to scan both horizontal and vertical barcodes. 6 Deck The deck is an enclosed work surface that is divided into 30 tracks. Samples, reagents and consumables are loaded onto the deck in carriers at specific track positions by the auto-loader. 5 Auto-load Tray The auto-load tray extends from the front of the deck with track markings to assist the carrier placement. The tray is divided into 30 tracks along with etched deck symbols. Samples, reagents and consumables are loaded onto the auto-load tray in the carrier at specific track positions. 4 Front Cover The front cover is a hinged, transparent cover that provides easy access to the deck. The cover is closed and locked during a run. 3 Power Switch for the Heater/Shaker This switch allows the operator to turn the heater/shaker on or off. 2 Power Switch for the VERSANT® SP Module The power switch turns the system on and off. 1 ? Interior of the System The front of the VERSANT® SP Module consists of the following six (6) components: Carriers Heater/shaker unit Magnetic Separator Waste Assembly Internal Swivel Arm Plate Handler (iSWAP) Pipetting Channels Select the colored callouts on the left to learn more about the interior of the system. Select the colored callouts on the left to learn more about the interior of the system. iSWAP The iSWAP transports the 2-ml DWP(s) and/or the 96 well PCR plate(s) to and from the required processing locations depending on the protocol selected. Carriers Each carrier occupies a designated track position on the deck of the instrument. There are removable carriers as well as a fixed position carrier. Heater/shaker Unit The SP Module houses one heater/shaker unit. Do not touch the heater/shaker. The heater/shaker may be hot and cause bodily injury when touched. Pipetting Channels There are eight pipetting channels that work in parallel for simultaneous transfer for liquids. Waste Assembly The waste assembly consists of the following: Liquid waste trough and tip ejection plate Liquid waste container Solid waste container Magnetic Separator The VERSANT® SP Module has a single magnetic separator that the DWP is temporarily positioned on during a protocol. ? System Carriers The deck of the VERSANT® SP module consists of carriers that are removable and a single carrier in a fixed position. Removable carriers include: Plate Carrier Tip Carrier Reagent Trough Carrier Control Carrier Sample Carrier The incubation and separation assembly is the only carrier in a fixed position. Removable carriers include: Plate Carrier, Tip Carrier, Reagent Trough Carrier, Control Carrier, Sample Carrier. The incubation and separation assembly is the only carrier in a fixed position. Select the numbered steps below to learn more about system carriers. Note: The plate carrier houses: Two deep-well plates, Up to two 96-well PCR plates, 300 microliter tip tray. The carrier has the capacity to hold two deep-well plates and up to two 96-well PCR plates depending on the protocol selected. Note: The VERSANT SP Module deck contains two tip carriers which can accomodate the following: 1000 microliter tips and 300 microliter tips. Note: The large trough carrier contains reagents in the following positions: position 1, lysis buffer. Position 2: wash one. Position 3: wash two. Position 4: wash three. Note: The small trough carrier contains reagents in the following positions: Position 1: Empty. Position 2: Empty. Position 3: Empty. Position 4: Magnetic Beads. Position 5: Elution Buffer. Note: The control carrier contains reagents used by the SP Module. The control carrier holds assay-specific reagents in the appropriate positions and Proteinase K in position 32. The control carrier is the same as a 32-position sample carrier, but it used with special adapters as the controls are 2 milliliter tubes. Note: The VERSANT SP Module deck contains two sample carriers which can accomodate 24 and 32 samples each. Note: The incubation and separation assemble is the only carrier that is fixed to the deck. The incubation and separation assembly contains the following: Heater/shaker unit, Magnetic separator, Two park positions. Incubation and Separation Assembly The incubation and separation assembly is the only carrier that is fixed to the deck. The incubation and separation assembly contains the following: Heater/shaker unit Magnetic separator Two park positions 7 Sample Carrier The VERSANT® SP Module deck contains two sample carriers which can accomodate 24 and 32 samples each. 6 Control Carrier The control carrier contains reagents used by the VERSANT® SP Module. The control carrier holds assay-specific reagents in the appropriate positions and Proteinase K in position 32. Note: The VERSANT® SP Module accommodates two sizes of reagent troughs- large(200 milliliter) and small (50milliliter). Note: The VERSANT® SP 1.0 kit contains the following in box 1: Lysis and Elution Buffer, Wash Buffer 1, 2 and 3 and Magnetic Beads. The SP 1.0 kit contains the following in box 2: Proteinase K, 1 tube and a barcode card. Note: The VERSANT ®SP 1.2 kit contains the following in box 1: Lysis and Elution Buffer, Wash Buffer 1, 2 and 3 and Magnetic Beads. The SP 1.2 kit contains the following in box 2: Proteinase K, 1 tube and a barcode card. SP 1.0 Reagents Kit The VERSANT® SP Module accommodates two sizes of reagent troughs- large(200mL) and small (50mL). Reagent Trough Sizes The VERSANT® SP Module accommodates two types of tips- 1000 and 300µL. Tip Volumes ? Software Overview Software Overview ? Verifying or Entering Kit Lot Information for Sample Reagents The VERSANT® kPCR Molecular System SP Module software allows the operator to verify and enter the kit lot information for sample reagents. Select the start button below to learn more about how to verify or enter kit lot information on the VERSANT® kPCR Molecular System SP Module. Start Verifying or Entering Kit Lot Information for Sample Reagents Note: The Versant KPCR Molecular System SP module software allows the operator to verify and enter the kit lot information for sample reagents. Select the start button below to learn more about how to verify or enter kit lot information on the VERSANT kPCR Nolecular System SP Module. Note: To verify and enter the kit lot information, perform the following actions in the software: Select the desired protocol in the main screen. Note: Verify that the kit information for the sample reagents is correct. If necessary, enter the lot specific information using the hand-held scanner. At the protocol tab, select Edit Lots. Note: The kit lot list window displays. Select the appropriate lot from the list, select view. Verify the information displayed is correct for the kit lot you plan to run. Note: The system returns to the kit lot list window. If the information is correct, select close to return to the main system window and start your run. If kit information is missing, select create. Starting a Run Before starting the run, ensure that all of the carriers are loaded in the correct positions on the Autoload tray. Select the Start button below to learn more about how to start a run on the VERSANT® SP Module. Start Starting A Run Note: Before starting the run, ensure that all of the carriers are loaded in the correct positions on the autoload tray. Select the start button below to learn more about how to start a run on the VERSANT SP Module. Note: Select the protocol tab in the main screen. Next, select the proper protocol from the list. Finally, select Run Protocol. Note: The run status window displays followed by the Empty the Solid Waste Container Window. When that appears, open the waste door located on the right side of the SP module. Remove the solid waste container. The Install Solid Waste Container window appears once the solid waste container has been removed. Empty the solid waste container and dispose of the waste as described in the operator manual. Reinstall the solid waste container and close the waste door. Note: Throughout the run, the Run Status window will display the progression of the run. The time remaining in the run will also appear at the bottom of the screen. If an error occurs, select Stop. Note: A confirmation window will display once Stop is selected. Select Yes to stop the run. ? The VERSANT® Sample Preparation reagents extract RNA or DNA from various types of patient samples depending on the protocol selected. Completing A Run At the end of a run, the VERSANT®SP Module defines the plate details and generates a run report. Select the button below on how to complete a run on the VERSANT® kPCR SP Module. Start Completing A Run Note: At the end of a run, the VERSANT SP Module defines the plate details and generates a run report. Select the start button below on how to complete a run on the VERSANT SP Module. Note: When the Run is complete, the software will audibly beep and then the Run Complete window displays in the software. When the run completes, verify that the autoload tray os clear and do the following to complete the run: Select Eject Carriers to end the run without viewing the run report file. The system closes the Run Complete dialog box, unloads all carriers, and ends the run. Select View/Print Report to preview the run report file and check results before ending the run. The system closes the Run Complete window, displays the Sample Preparation Run Report, unloads all carriers and ends the run. Select Finalize Run to eject carriers, finish the run, and return to the main screen. Select Eluate Storage to use the Eluate Storage function. Refer to Eluate Storage in the operator manual for more information. ? SP Module Maintenance The VERSANT® SP Module has four daily and weekly maintenance tasks. Note: The steps listed show the operator how to perform access maintenance on the SP module. If both daily and weekly maintenance are required, select weekly instead of daily maintenance. Note: To clean the deck, Spray Microcide SQ on a clean, lint-free laboratory wipe and wipe to clean both the deck and the tip eject plate. Rinse using deionized water and follow by spraying with a 70% ethanol solution. Do not directly spray the SP Module deck with Microcide SQ. Microcide SQ can damage electrical boards and connectors. Note: Remove and clean the solid waste container. Line the solid waste container with a new biohazard bag and place the solid waste container back on the system. Dispose of the biohazard bag according to your laboratory and local regulatory protocols. Ensure the biohazard bag us laying flat against the container. Note: When the Daily or Weekly Maintenance - successful dialog box displays, select OK, which verifies that the process has completed successfully. If the maintenance procedure is aborted, this dialog box will not display. The system displays a warning indicating that if you stop the maintenance before it is complete, you must turn the VERSANT SP Module off for 10 seconds, then turn it back on. Once the system has rebooted, rerun the maintenance procedure. Verify Completion When the Daily or Weekly Maintenance - successful dialog box displays, select OK, which verifies that the process has completed successfully. If the maintenance procedure is aborted, this dialog box will not display. The system displays a warning indicating that if you stop the maintenance procedure before it is complete, you must turn the SP Module off for 10 seconds, then turn it back on. Depending on the protocol selected, there may be additional assay specific reagents that may also need to be removed. Any remaining reagents that you wish to keep should be capped and returned to the recommended temperature conditions. Discard all used reagents and troughs (liquid and solid waste) according to the practices of your institution and in compliance with all local and regulatory requirements. Note: Discard the 96-well PCR and/or deep-well plate into the biohazard waste container in accordance with laboratory and local regulatory protocols. Note: Remove and clean the solid waste container. Line the solid waste container with a new biohazard bag and place the solid waste container back onto the system. Dispose of the old biohazard bag according to laboratory and local regulatory protocols.
